Mechanisms and applications of disulfide bond formation

The research undertaken in this project aims to develop efficient systems for the production of disulfide bond containing proteins in the cytoplasm of E.coli and apply these for structural and functional characterization of disulfide bond containing proteins of scientific or biomedical importance. Our system is already approaching levels of CHO for the production of aglycosylated full length antibodies and we have recently used it to gain new structural insights into the mechanisms of lipid metabolism and protein folding as well as the organization of basement membranes.
