My postdoc research project focuses on the Cistromic and Proteomic characterization of human transcription factors (TFs) in cancer. We have established around 200 TF overexpression prostate cancer cell lines. We have further performed the Chromatin Immunoprecipitation Sequencing (ChIP-seq) of these TFs to study the protein-DNA interaction, and in cooperation with University of Helsinki we did the Affinity Purification Mass Spectrometry (AP-MS) of these TFs to study the protein-protein interaction (PPI). With the bioinformatic analysis method, we expect to know how the TFs regulate 3D chromatin reprogramming in prostate cancer, how the TFs regulate prostate cancer risk genes. Combined with genome-wide association studies (GWAS) catalog of prostate cancer data, we can find the TFs regulated prostate cancer risk SNPs. Combined with the Finnish cohort of human prostate cancer patients, we can find potential prostate cancer therapy target. These results will be a major part of my project and may facilitate the development of genetic markers for improving prostate cancer risk prediction and developing therapeutic strategies against prostate cancer in men.
