The Jianan Lab of Nanophotonic Biosensors

Because proteins cannot be amplified, current protein analysis and sequencing relies on mass spectroscopy that generally needs 1 billion copies of the proteins. The low sensitivity of mass spectroscopy leads to a huge challenge in detection of low-abundance proteins in human biofluids and thus spurs extensive research interests in single-molecule protein sequencing. Jian-an developed a plasmonic nanopore system by electro-plasmonic trapping of a gold nanoparticle in a gold nanopore to generate single SERS hot spot. The nanopore was so sensitive and localized that it not only distinguished single DNA bases in single single-strand DNA [1] but also discriminated single amino acid residue in single polypeptide [2]. In addition to the sub-molecular resolution, the plasmonic nanopre system has demonstrated single-molecule detection of 10 different types of amino acids by their Raman spectra [2], which has paved a completely new way to single-molecule protein sequencing. To apply the nanopore system to single-molecule sequencing, a microfluidic method will be integrated to stretch the DNA or protein molecules and slow down their passage through the nanopore.

Wearable non-invasive molecular sensing technology will be an essential part for future personalized medicine. Current non-invasive wearable devices monitor heart rate and other physical, kinetic values such as temperature and body gestures, steps and location. It would be of tremendous added value if they could identify and quantify health factors that take part in control of human physiology, such as sweat glucose molecules that reflects blood glucose level. Failure to control blood glucose increases Body Mass Index, resulting in tremendous health risks in current society. This project aims to develop a wearable optical sensor on skin by integrating an electro-plasmonic chip [1] into an optical watch that together allows controllable sweat stimulation and on-watch continuous detection of the sweat glucose. When fully implemented, we plan to explore integration of an spectrometer into the watch and extend the lab-in-a-watch sensor to detect extracellular vesicles in sweat.

Single-cell analysis provides important information of biology that are masked in analysis of cell populations and tissues. Current single-cell analysis techniques allow cell analysis at the genomic, transcriptomic and proteomic levels but have to lyse the cells to extract the cytosolic contents. Thus, it hampers real-time monitoring of intracellular dynamic transformations, such as differentiation and gene expression of single living cells, and, more importantly, interconnection between living cells with time. What is worse, the amount of extracted molecules from single cell were too little to be analyzed by conventional methods such as mass spectrometery.

The research goal of this topic is to develop optical biosensing methods using surface-enhanced Raman spectroscopy (SERS) as the sensing function for single-molecule and real-time single-cell analysis. A hollow nanoelectrode system is used to electroporate cell membrane, after which molecules or nanoparticles can be delivered into cytosol.[1] Alternatively, It can extract and sample intracellular content from specific cell into the microfluidic channel by electrical bias.[2] The extract can be detected by the plasmonic nanopores for single-molecule SERS characterization of the intracellular content.[3] By cycling sampling and detection, the integrated system will find broad applications for different cellular systems, such as cancer cells response to drugs.
